Chemistry: Elusive mechanism of widely used click reaction revealed

Apr. 4, 2013 ? Scientists at The Scripps Research Institute (TSRI) have illuminated the mechanism at the heart of one of the most useful processes in modern chemistry. A reaction that is robust and easy to perform, it is widely employed to synthesize new pharmaceuticals, biological probes, new materials and other products. But precisely how it works had been unclear since its invention at TSRI more than a decade ago.

"These new findings allow us to exert greater control of the reaction and make it faster and more efficient under the most challenging conditions," said chemist Valery Fokin, an associate professor at TSRI, who was principal investigator for the new study. "The reaction-tracking techniques we developed here also can be applied to the study of other complex processes, both chemical and biological."

The report, which sheds light on the reaction known as copper-catalyzed azide-alkyne cycloaddition (CuAAC), appears on April 4, 2013 in Science Express, the advance online edition of the journal Science, and in the April 18, 2013 issue of the journal.

Classic Click Reaction

Fokin and his laboratory, and the laboratory of K. Barry Sharpless, a Nobel laureate and the W.M. Keck Professor of Chemistry at TSRI, reported the discovery of the CuAAC reaction in 2002. Danish researchers independently reported a similar reaction in the same year. The reaction involves the use of copper compounds to catalyze the linkage of two functional groups, a nitrogen-containing azide and a hydrocarbon alkyne, to make a stable five-membered heterocycle, 1,2,3-triazole. Azides and alkynes are small functional groups that can be easily introduced into a wide variety of structures using chemical or biological methods without interfering with normal biological processes.

The experimental simplicity and reliable performance of CuAAC under virtually all conditions, including in water and in the presence of oxygen, has made it a "go-to" method whenever covalent stitching of small human-made molecules or large biopolymers is needed, exemplified by protein and nucleic acid labeling, in vitro and in vivo imaging, drug synthesis and the forging of complex molecular architectures with surgical precision.

"Despite its many uses, the nature of the copper-containing reactive intermediates that are involved in the catalysis had not been well understood, in large part due to the promiscuous nature of copper, which rapidly engages in dynamic interactions with other molecules," said Fokin.

Previous studies had hinted that in the swirl of short-lived bondings and partings that occur during a given CuAAC reaction, not one but two copper-containing catalytic units -- "copper centers" -- are needed to help build the new triazole structure. To confirm this, Fokin and two of his graduate students, Brady Worrell and Jamal Malik, tried to reproduce key steps of the CuAAC catalytic cycle with either one or two copper atoms available. Analysis of the reaction course by tracking the heat given off by each reaction as well as product yield indicated whether it worked efficiently. "By monitoring the reaction in real time, we showed that both copper atoms are needed and established the involvement of copper-containing intermediates that could not be isolated or directly observed," said Worrell, who was the paper's first author.

In a second set of experiments, Worrell, Malik and Fokin introduced a pure isotope of copper -- which differs slightly in mass from the isotope blend found in natural copper -- as one of the two copper centers so that they could track their respective fates during the reaction. "We hypothesized that the two copper centers would have distinct roles, but found unexpectedly that their functions during key steps in the reaction are effectively interchangeable," said Malik.

New Linkages

The research reveals the popular CuAAC reaction in unprecedented detail. In addition to the fundamental insights into the chemistry of copper and its interactions with organic molecules, the techniques will lead to better understanding of many chemical and biological processes involving copper. The current study also enables development of new reactions that exploit weak interactions of copper catalysts with carbon-carbon triple bonds. In fact, based on the new findings, Fokin and his team have begun to devise new reactions in which one copper center can be replaced with a different element, to pursue complementary biocompatible and efficient techniques.

(AP) ? A middle-aged man who transported poultry for a living and another unidentified person have died from a new strain of bird flu, bringing the death toll to five among 14 confirmed cases in China, the government and state media reported Thursday.

The 48-year-old man, who died in Shanghai, is one of several among the infected believed to have had direct contact with fowl. Until recently, the virus, called H7N9, was not known to infect humans.

The official Xinhua News Agency did not identify the fifth fatality, but said that person also died in Shanghai on Wednesday.

It said the Ministry of Agriculture confirmed on Thursday that the H7N9 virus had been detected in pigeons at a market selling agricultural products in Shanghai.

It is not known how people are becoming sick with the virus, and health officials and scientists caution that there are no indications it can be transmitted from one person to another. Scientists who have studied the virus's genetic sequence said this week that the virus may have mutated, spreading more easily to other animals and potentially posing a bigger threat to humans.

Guidelines issued Wednesday by the national health agency identify butchers, breeders and sellers of poultry, and those in the meat processing industry as at higher risk.

Experts only identified the first cases on Sunday. Some among the 14 confirmed cases fell ill several weeks ago but only now are being classified as having H7N9.

Xinhua said six cases have been confirmed in Shanghai, four in Jiangsu, three in Zhejiang and one in Anhui.

BillPin Acquires Obopay's BillMonk Bill-Splitting Service

BillPin LogoAfter some years of languishing, BillMonk’s service and user base will be taken over by a young Singapore startup, BillPin, which provides a similar bill-splitting service. BillPin pitches itself as “Mint for groups”, and offers a way for people to track how much money they owe each other, and is barely over six months old. It has plenty of competitors in the bill-splitting space that includes Splitwise, Billsup, Spotme, Divvyit, Splitmybill.ie, and scooping up BillMonk’s user base might provide it a firm leg up in the fight for user acquisition. BillMonk was founded in 2005 and was acquired by Indian mobile money provider, Obopay in 2007. It’s one of the older players in the space, and seems to have suffered from neglect for a few years. Some of its competitors have tried to capitalize on that. Splitwise launched a campaign to bring BillMonk users onboard last year. Funnily, BillPin too had a similar service launched in October last year. BillPin’s co-founder, Darius Cheung (whose previous startup, tenCube WaveSecure, was acquired by McAfee in 2010), acknowledged the older firm’s state of neglect.?”Not much improvement was added to BillMonk at all over the past seven years, and in fact BillMonk has suffered significant down time in the last 6 months or so,” he said. Of the migration service BillPin created, he said: “It was launched to catch BillMonk users who were frustrated with its downtime, but we didn’t know then we were going to inherit its entire user base.” Still, BillMonk continued to add users over its lifetime, and steadily at that, he said. As a result, bringing BillMonk’s user base onboard will give BillPin “easily 50 times” more users. He wouldn’t say how many that is, but according to reports, BillPin has a small base of 5,000 or so users. He made it clear that this move was an acquisition of the larger service’s user base, but none of BillMonk’s engineers will come over from Obopay. BillPin has five employees in Singapore and one in India. It’s bootstrapped so far, but Cheung is currently hoping to raise an angel round. Obopay started in 2005, and provides its technology to corporate clients like Nokia and Societe Generale, and telcos like Warid Telecom in Uganda to allow them to offer their own branded mobile money transfer services. Using new technology and new approaches, researchers at Lund University in Sweden hope to be able to explain why people suffer vision loss in eye diseases such as retinal detachment and glaucoma.

Research on diseases of the eye such as retinal detachment and glaucoma has until now focused on the biochemical process that takes place in the eye in connection with the diseases.

Fredrik Ghosh and Linna Taylor have concentrated instead on attempting to understand what happens on a biomechanical level in the diseases and have produced results that have drawn a lot of interest from experts.

"We have not previously understood the mechanisms behind glaucoma and retinal detachment, but we knew that these diseases had a strong mechanical component. Our findings could form an initial explanation as to why we develop these diseases", said eye researchers Fredrik Ghosh and Linna Taylor.

Using new technology, the eye researchers at the Department of Clinical Sciences in Lund, in collaboration with researchers at the Department of Biology at Lund University, have developed a method to investigate the importance of the biomechanical environment within the central nervous system.

For their studies, they grow retinal tissue from adult pigs in a stretched state similar to the normal mechanical state present in the living eye. Compared with unstretched tissue, which in cultures dies after a few days when the retina's mechanical balance is disturbed, studies can now be performed for up to ten days in retina with a well-preserved structure and significantly higher cell survival.

"This gives us new tools to understand in a more concrete manner how biomechanical factors in the central nervous system influence the health of cells when we are healthy and when we suffer from diseases. This will not only have major importance for our understanding of how diseases come about in the central nervous system, but also for future disease treatment", said the researchers.

The central nervous system, which includes the brain, spinal cord and retinas, is a complicated organ, especially in terms of structure. The entire system is under the mechanical influence of fluid pressure, among other factors. The new data from the group in Lund indicates that when the biomechanical balance is disturbed, as happens in retinal detachment and glaucoma, the normal function of the retina is lost, resulting in serious sight impairment or blindness.

(AP) ? The U.N. General Assembly overwhelmingly approved the first international treaty regulating the multibillion-dollar global arms trade Tuesday, after a more than decade-long campaign to keep weapons from falling into the hands of terrorists, warlords, organized crime figures and human rights violators.

Loud cheers erupted in the assembly chamber as the electronic board flashed the final vote: 154 in favor, 3 against and 23 abstentions.

"This is a victory for the world's people," U.N. Secretary-General Ban Ki-moon said. "The Arms Trade Treaty will make it more difficult for deadly weapons to be diverted into the illicit market. ... It will be a powerful new tool in our efforts to prevent grave human rights abuses or violations of international humanitarian law."

The United States, the world's biggest arms exporter, voted yes.

Iran, North Korea and Syria ? all facing arms embargoes ? cast the only no votes. They argued, among other things, that the agreement favors major arms suppliers like the U.S. over importers that need weapons for self-defense.

Russia and China, which are also major arms exporters, abstained along with India and Indonesia, while nuclear-armed Pakistan voted in favor. Many Arab countries, including Egypt, Saudi Arabia, Sudan and Qatar, abstained, while Lebanon voted yes.

Never before has there been a treaty regulating the global arms trade, which is estimated to be worth $60 billion today and which Amnesty International predicts will exceed $100 billion in the next four years.

"Today's victory shows that ordinary people who care about protecting human rights can fight back to stop the gun lobby dead in its tracks, helping to save countless lives," said Frank Jannuzi, deputy executive director of Amnesty International USA.

"The voices of reason triumphed over skeptics, treaty opponents and dealers in death to establish a revolutionary treaty that constitutes a major step toward keeping assault rifles, rocket-propelled grenades and other weapons out of the hands of despots and warlords who use them to kill and maim civilians, recruit child soldiers and commit other serious abuses."

What impact the treaty will actually have remains to be seen. It will take effect 90 days after 50 countries ratify it, and a lot will depend on which ones ratify and which ones don't, and how stringently it is implemented.

As for its chances of being ratified by the U.S., the powerful National Rifle Association has vehemently opposed it, and it is likely to face stiff resistance from conservatives in the Senate, where it needs two-thirds to win ratification.

Secretary of State John Kerry called it a "strong, effective and implementable" treaty and stressed that it applies only to international deals and "reaffirms the sovereign right of any state to regulate arms within its territory."

The treaty prohibits countries that ratify it from exporting conventional weapons if they violate arms embargoes, or if they promote acts of genocide, crimes against humanity or war crimes, or if they could be used in attacks against civilians or schools and hospitals.

Countries must also evaluate whether the weapons would be used by terrorists or organized crime or would undermine peace and security. They must take measures to prevent the weapons from being diverted to the black market.

The treaty covers battle tanks, armored combat vehicles, large-caliber artillery systems, combat aircraft, attack helicopters, warships, missiles and missile launchers, and small arms and light weapons.

Enforcement is left up to the nations that ratify it. The pact requires these countries to assist each other in investigating and prosecuting violations.

"The treaty is a noble gesture that may over time acquire the kind of precedence or enforcement that would give it meaning," said Anthony Cordesman, an analyst at the Center for Strategic and International Studies in Washington. "At this point it is more a declaration of principles ? and the arms trade is an area where many people don't have principles."

Supporters of the treaty agreed that it is just a first step and that it must be followed by a campaign for implementation.

"The hard work starts now," said Juan Manuel Gomez Robledo, Mexico's vice minister for multilateral affairs.

Australian Ambassador Peter Woolcott, who chaired the negotiations, said the U.S. "played a hugely constructive role" in pushing the treaty through the United Nations.

"Obviously, as the world's largest exporter, it would be unfortunate for the Arms Trade Treaty if the U.S. didn't sign it, but obviously it's a sovereign decision for them," he said.

Hopes for adoption of the treaty by consensus instead of a vote were dashed last July when the U.S. said it needed more time to consider it.

At the end of the final negotiating conference last week, Iran, North Korea and Syria blocked another attempt at consensus. Over those countries' objections, the treaty's supporters decided to put it to a vote in the General Assembly.

Proponents of the treaty said it could make it much harder for regimes committing human rights violations to acquire arms, in conflicts such as the brutal civil war in Syria.

"The treaty's prohibition section, if it were in force today, would prohibit the ongoing supply of weapons and parts and components to the Assad regime in Syria," said Daryl Kimball, executive director of the independent Washington-based Arms Control Association.
